1.     Ph. D. Thesis on Oxidative Enzymatic Processes (PIF contract)

European Project (Horizon 2020) entitled: Expanding the industrial use of Robust Oxidative Bioctalysts for the conversion and production of alcohols (ROBOX).

ROBOX Project Objective: In order to achieve the widening of industrial application of enzymatic biooxidation processes, ROBOX will demonstrate the techno-economic viability of biotransformations of four types of oxidative enzymes:

Cytochrome P450 monooxygenases (P450s)

Baeyer-Villiger MonoOxygenase (BVMOs)

Alcohol DeHydrogenases (ADHs)

Alcohol OXidases (AOXs)

The ROBOX consortium aims to make biocatalytic alternatives, based on probably the three most important types of enzymatic oxidations (alcohol oxidation, C-H activation and lactonization by monooxygenases) with reduced environmental footprint accessible to large scale chemical synthesis and other industrial conversions. This will be done by the development and demonstration of 11 industrially relevant reactions catalyzed by representatives of four different types of oxidative enzymes, while concomitantly consolidating the corresponding experimental and computational methods into a generic platform of technologies for fast and accurate enzyme activity determination required for enzyme optimization, production and application in a biocatalytic process. This approach will enable the design of highly efficient biocatalysts with an adapted range of substrates of immediate relevance for industrially important conversions.

Ph. D. Thesis on Multi Enzymatic Synthesis Processes

Spanish  Project  (CTQ2014-53114-R (MINECO)) entitled: Intensification of Multi Enzymatic Processes (IMEP).

IMEP Project Objective: to apply the concept of process intensification to multi-enzymatic biocatalysis to develop enzymatic processes with industrial interest and strengthen the biocatalysis viability as an alternative to catalytic technology. In particular, the synthesis of two types of compounds is intended: chiral amines and β-aminopolyols. In both cases, multi-enzymatic systems are required and the target products have great industrial interest.

The project objective is to intensify the following biotransformations:

- Multi-enzyme synthesis of chiral amines catalyzed by immobilized ω-transaminase (TA) and pyruvate decarboxylase (PDC): 1-phenyl-ethyl-amine and 3-amino-1-phenyl-butane (a precursor of antihypertensive dilevalol).

-Multi-enzyme synthesis of β-aminopolyols catalyzed by immobilized chloroperoxydase (CPO) and different aldolases to render various β-aminopolyols. These compounds are precursors of iminocyclitols with potential biological activity. In particular, D-fagomine, a target compound for this biotransformation, is an iminosugar with anti-hyperglycemic and antimicrobial properties.

In summary, the processes intensification will be carried out in the following items:

Biocatalyst engineering: Preparation of robust biocatalysts through enzyme immobilization/ stabilization. Immobilized derivatives with high enzymatic load will be prepared to increase enzymatic reaction rates and catalyst productivity (g product / g of biocatalyst). Also, co-immobilization of different enzymes in the same support will be used as an alternative to immobilize each enzyme separately. Immobilization will be performed by different conventional methods and, also, immobilization on magnetic nanoparticles will be assessed. In the latter case, the synthesis would proceed trough homogeneous catalysis without diffusional limitations.

Multi-enzymatic reactions engineering: The main objective is to optimize the operating conditions to increase the concentrations of substrate, maintaining the activity of the biocatalyst (stability).

Reactor engineering: Alternative configurations for simultaneous reactions in one reactor (one-pot) and for consecutive reactions (in reactors in series or in the same reactor) will be analyzed in order to optimize kinetics and productivity. When nano-biocatalysts were used, the integration of reaction/separation processes is intended.
